Twitter bootstrap is, without a doubt, one of the best things to help the prototyping project. I am currently redoing this site, and it is being done in Twitter Bootstrap with HTML5Boilerplate backing it up. One of the biggest problems that TB created is that, since it was so easy to use, we suddenly saw a boon of sites that all looked the same, or super similar - enough that you would notice.
